Get m6A and inosine mapping from the same samples, in addition to RNA seq. Correlate changes between modification types and find out which genes are up or downregulated in test and control conditions, and how the gene expression levels relate to RNA modification status. Identify equally expressed yet differentially modified genes to understand how the epitranscriptome modulates the proteome.

Relative Quantitation

Compare relative RNA modification levels.

Only a subset of RNA transcripts of a given gene contains RNA modifications to fine-tune function. The EpiPlex assay includes spike-in controls as normalization standards to infer the modification stoichiometry of each gene. Normalizing the assay signals in this way also reduces assay variability and enables data comparisons across operators and experiment batches.

Tissue biopsies often contain less than 500 ng of total RNA, challenging many RNA sequencing methods. With the EpiPlex Duo-Mod Service, you can obtain exceptional data quality with as little as 20 ng poly(A)-selected RNA or 250 ng total RNA.

Input requirements 250 ng total RNA (RIN ≥ 7), 20 ng poly(A)+ RNA (DV200 ≥ 90), 20 mg fresh frozen tissue or 500 ng total RNA from FFPE (DV200 ≥ 30).

Sample storage Suspend RNA in ethanol or RNA-free ultra-pure water and store at -80°C. Avoid repeated freezing and thawing.
Shipping method Ship the RNA sample in a 1.5 ml Eppendorf tube, sealed with sealing film. Shipments are generally recommended to contain 5-10 pounds of dry ice per 24 hours.
